Why don’t you use a probe directly in a reference sample to control the temperature during freezing in the EF600?
The EF600 uses a probe in the sample plate rather than in a reference sample because the design of the plate and the responsiveness of the Cryocooler means that that there are no significant temperature lags between the plate and the samples. Maintaining a fixed control probe ensures reproducibility of the protocols.
Furthermore unless ice nucleation occurred simultaneously in the reference sample and all other samples, which is unlikely, the sample to sample variation would be significantly worsened.
